New Approaches for Levodopa Treatment in Parkinson’s Disease
Parkinson’s disease (PD) is characterized by degeneration of dopaminergic cells, which results in dopamine depletion. Levodopa is the most effective symptomatic treatment, however, disease progression along with the unfavorable pharmacokinetics of levodopa makes the disease increasingly difficult to treat with time.

Development of Substance P 1–7 Related Peptides and Peptidomimetics: Targeting Neuropathic Pain
The aim of this thesis was to design and synthesise SP1–7 related peptides and peptidomimetics, to be used as research tools to study the SP1–7 system, and to serve as drug leads in the neuropathic pain area.

Clinical Consequences of Axonal Injury in Traumatic Brain Injury
The aim of this thesis was to study clinical consequences of axonal injury, its detection and pathological features, and potential link to neurodegeneration in severe TBI patients treated at the neurointensive care unit at Uppsala University Hospital.

Architecture and function of the insulin granule secretion machinery
In this thesis, high resolution TIRF microscopy was used to identify rate limiting steps for insulin granule exocytosis in health and in type-2 diabetes, and to understand these steps at the molecular level. It is shown that granule docking is critical for sustained insulin secretion. In β-cells from type-2 diabetic donors, docking is compromised and no longer responsive to glucose.

Birth asphyxia: Fetal scalp blood sampling and risk factors for hypoxic ischemic encephalopathy
Preventing birth asphyxia is a major challenge in delivery care. The aims of this thesis were to evaluate fetal scalp blood sampling (FBS) and explore risk factors for moderate to severe neonatal hypoxic ischemic encephalopathy (HIE).

Environmental Factors of Importance in Myasthenia Gravis: Emphasis on Physical Activity
In this thesis the occurrence of lifestyle related environmental factors in Myasthenia Gravis (MG) patients were investigated. MG epidemiology, subgroup differences and the presence of lifestyle related risks in MG patients were evaluated in Jönköping county in Southern Sweden. The findings were compared to a similar evaluation in a region of Estonia.

Understanding the genetic basis of complex traits
The aim of this thesis is to evaluate the genetic basis of complex traits by exploring available genomic resources and analytical approaches.

Targeting allelic loss in colorectal cancer
This thesis explores the consequences of allelic loss affecting tumor suppressor genes and passenger genes in colorectal cancer (CRC), aiming to identify vulnerabilities that can be exploited for therapy.

Structural Determination of Drug Metabolites from Doping Classed Compounds Using Mass Spectrometry
Doping control in equine sports is important for a fair competition, but also to ensure the integrity of the betting system, as well as for animal welfare reasons. To detect the use of illicit compounds, screening for the parent compound is common. However, by using a metabolite as the analytical target instead, the detection time can be prolonged.

Pericyte Influence on Drug Delivery Across the Blood-Brain Barrier: Implications for Therapy of Neurodegenerative Diseases
The aim of the thesis was to examine the role of pericytes for small-molecular drug transport across the blood-brain barrier (BBB), by providing a closer insight into different aspects of transport in a pericyte-deficient state.

Discrepancy between pregnancy dating methods – correlates and outcomes
With the introduction of obstetric ultrasound there has been a shift from last menstrual period-based to ultrasound-based estimation of gestational age. The choice of the method and timing of pregnancy dating is important because it can affect dating precision and perinatal outcomes.

Colorectal and appendiceal peritoneal metastases: From population studies to genetics
In this thesis, colorectal and appendiceal PM were studied from a wide population-based perspective down to the detailed perspectives of histopathology and genetics, with the aim of further contributing to prolonged survival.

Cervical Radiculopathy: Studies on Pain Analysis and Treatment
In patients with cervical DDD and radiculopathy both the pain drawing test and the VAS are validated tools to interpret the patients’ pain. Preoperative mental distress affects long-term outcome much more than the allocated treatment, ADR or fusion surgery in patients with cervical radiculopathy.

Second Victims in Swedish Obstetrics
The term “second victim” implies that healthcare providers can be pro-foundly affected by severe events in which a patient is badly injured or dies. The patient is the first victim. This thesis investigates the magnitude, riskfactors and consequences of becoming a second victim in Swedish delivery care.

Smokeless Tobacco (Snus) and Cardiovascular Disease: Associations with Heart Failure and Prognosis after Myocardial Infarction
The aim of this thesis was to investigate if snus is associated with development of heart failure and the prognosis after myocardial infarction. We used data from Swedish cohort studies and the national quality register for myocardial infarctions (SWEDEHEART), with linkages to national registers.

Determination of the Environmental Fate of Drug Substances and the Matrix Effects of Complex Samples in SFC/ESI-MS
Awareness of the potential problems caused by drug compounds in the environment has increased over the last decade, both among researchers and with the public. This thesis describes the development of analytical methods and their application to wetlands constructed for purification of wastewater from e.g. drug compounds.

Stress, Drugs and Neuroscience: Neurobiological Effects of Social Stressors and Drug Exposure in Young and Adolescent Rats
Experiences early in life or during adolescence modulate neuronal networks in the immature brain and consequently lay the foundation for future susceptibility or resilience towards psychiatric disorders. The objective in this thesis is to understand, in part, how the surrounding environment shapes the brain of a young individual.
